Because of its widespread use, sex attractant biosynthesis has been the subject of numerous reviews. Yet, in recent years it has become apparent that our basic knowledge of the genes and proteins involved in biosynthesis is still fragmentary. In this review, the historical context and experimental advances in this field are examined. Two models, sex pheromone glands and the olfactory system of Drosophila, are described. The production of various types of compounds, including linear, linear-branched, and branched acetates, methyl ketones, alcohols, esters, phenols, and steroids, by both these model systems are summarized, and then examples of other insect orders are described. A special emphasis is placed on the latest discoveries about the insect P450s (cytochromes P450) and their enzymatic specificity, reaction mechanisms, and putative physiological functions.
